Strategies For Immunogen Design

There are many considerations when inducing an immunized animal to produce appropriate target-specific antibodies. An antigen must be prepared and delivered in a form and manner that maximizes the production of a specific immune response. We consider numerous variables when designing our immunogens.

 

We use many design tools to select an optimal antigen sequence.

Each program helps create a thorough map of the protein and helps accurately predict the general characteristics such as hydrophobic, hydrophilic regions, secondary structure, transmembrane regions, flexible regions, and surface probability.

 

Homology Analysis: Species homology and family protein homology are analyzed. 

 

Epitope Selection: Optimal antigenic epitopes should be flexible, hydrophilic, located on the protein's surface.

 

Continuous Epitopes or Discontinuous Epitopes Analysis: We select continuous epitopes for most research antibody projects. Discontinuous epitopes are considered only for functional antibody projects.

 

N-terminus or C-terminus sequence: We preferentially selected suitable epitopes in the N-terminal or C-terminal region. 

 

Sequence Length: We recommend that peptide sequences of 8-20 amino acids or recombinant protein of 200-300 amino acids. 

 

Immunogen Purity: The purity of synthetic amino acids should be more than 85%, and the purity of expressed protein should be more than 80%. 

 

Selection for Carrier Proteins and Expression Systems: KLH, OVA, or BSA as carrier protein will are determined according to the project's requirements, and expression systems E.coli or 293T/F will be selected according to the needs of the project. 

 

Peptide Modifications Analysis: Glycosylation sites are avoided.
